Control of Pyridoxine Biosynthesis in Escherichia Coli.

چکیده
Dempsey, Walter B. (University of Florida, Gainesville). Control of pyridoxine biosynthesis in Escherichia coli J. Bacteriol. 90:431-437. 1965.-The total pyridoxine in a culture of exponentially growing Escherichia coli was 3.6 x 10(-10) moles per mg of dry cells. One-fourth of this total was present in the medium, and was at least 90% pyridoxal 5'-phosphate. Both pyridoxol and pyridoxal, when present initially at 6 x 10(-7)m, substituted entirely for de novo synthesis of pyridoxine. The other four forms of the pyridoxine group were ineffective at this concentration. Pyridoxine biosynthesis in exponentially growing cultures of E. coli was immediately stopped by adding pyridoxol to the medium to a final concentration of 4 x 10(-7)m. Amino acid auxotrophs of E. coli suspended in minimal medium without the required amino acid produced extracellular pyridoxine for several hours at an average rate of 1.3 x 10(-10) moles per hr per mg of dry cells. The rate of production of extracellular pyridoxine by a threonine-starved threonine auxotroph was not altered by growing the culture before starvation in media containing 6 x 10(-5)m pyridoxol. The extracellular form of pyridoxine produced in these starvations required hydrolysis to be detected in the bioassay used in these measurements. The total findings suggest that pyridoxine biosynthesis is controlled by a rapidly acting mechanism but not by repression.

Isolation of PDX2, a second novel gene in the pyridoxine biosynthesis pathway of eukaryotes, archaebacteria, and a subset of eubacteria.
In this paper we describe the isolation of a second gene in the newly identified pyridoxine biosynthesis pathway of archaebacteria, some eubacteria, fungi, and plants. Although pyridoxine biosynthesis has been thoroughly examined in Escherichia coli, recent characterization of the Cercospora nicotianae biosynthesis gene PDX1 led to the discovery that most organisms contain a pyridoxine synthesi...
متن کاملEffect of Concomitant Lycopene Biosynthesis on CoQ10 Accumulation in Transformed Escherichia coli Strains
CoQ10 and lycopene are isoprenoid compounds with nutraceutical and pharmaceutical benefits. In this study, the effect of concomitant lycopene biosynthesis on CoQ10 accumulation in transformed Escherichia coli DH5α was studied. A lycopene production pathway including geranylgeranyl diphosphate synthase (crtE), phytoene synthase (crtB), and phytoene desaturase (crtI) from Erwinia herbicola was co...
متن کاملA highly conserved sequence is a novel gene involved in de novo vitamin B6 biosynthesis.
The Cercospora nicotianae SOR1 (singlet oxygen resistance) gene was identified previously as a gene involved in resistance of this fungus to singlet-oxygen-generating phototoxins. Although homologues to SOR1 occur in organisms in four kingdoms and encode one of the most highly conserved proteins yet identified, the precise function of this protein has, until now, remained unknown. We show that ...
متن کاملPyridoxine-requiring mutants of Escherichia coli: glycolaldehyde dehydrogenase is not coded for by the pdxB gene.
Twenty-seven independent pyridoxineless mutants belonging to genetic linkage group I were assayed for glycolaldehyde dehydrogenase. Some mutants lacked enzyme activity entirely, and others showed activity ranging from very low to wild-type levels. Reversion to pyridoxine independence usually had no effect upon this activity. Transfer of the pyridoxine genes to a common host that had wild-type l...
متن کاملThe Conversion of Pyridoxine Phosphate into Pyridoxal Phosphate in Escherichia Coli.
1. Evidence is presented for the presence of pyridoxine phosphate oxidase in aqueous extracts of Escherichia coli. Some comparison is made with pyridoxamine phosphate oxidase. 2. Isoniazid and iproniazid were found to combine with pyridoxal phosphate, but isoniazid did not combine with either pyridoxamine phosphate or pyridoxine phosphate. Both oxidase activities were somewhat inhibited by benz...
